The key enzyme is Prolyl Endopeptidase (PEP) — a proline-specific protease that cleaves the proline-rich peptide bonds that are the specific bonds in gluten/gliadin that are resistant to human digestive enzymes (most human proteases cannot efficiently cleave proline-adjacent bonds). PEP specifically targets the T-cell epitopes in gliadin that trigger immune reactions in susceptible individuals.
